Replicative senescence raises a handful of sensible questions. This page answers them in order, starting with the fundamentals and moving to applications.

Documentation accompanying research peptides usually includes a certificate of analysis listing the batch number, purity figure, and test methods applied. Buyers comparing suppliers should check whether the reported purity refers to chromatographic area or to a mass-balance calculation, because the two are not equivalent. Counter-ion content, residual solvents, and water content are sometimes omitted from such certificates even though they affect the actual peptide mass present. Independent verification through a second laboratory is the most direct way to confirm that a supplied material matches its label.
Purity assessment of peptide reagents normally relies on reversed-phase high-performance liquid chromatography. A gradient of acetonitrile in water with trifluoroacetic acid, paired with a C18 stationary phase, separates the target tetrapeptide from truncated sequences and deletion analogues. Detection at 214 nanometres exploits absorbance of the peptide backbone, since the molecule contains no aromatic residue. Results are expressed as a percentage of total peak area. Values above ninety-five percent are typical for research-grade material, although reporting conventions vary between suppliers.
Identity confirmation uses mass spectrometry, usually electrospray ionisation in positive mode or matrix-assisted laser desorption. The protonated free peptide produces a signal near three hundred and ninety-one daltons, while salt adducts shift that value slightly. Tandem mass spectrometry or amino acid analysis supplies sequence-level confirmation, which a single mass measurement cannot. Peptides containing aspartate can cyclise into succinimide intermediates that hydrolyse to isoaspartate isomers of identical mass. Because those isomers are difficult to separate chromatographically, identity and purity results carry more weight when interpreted together.
Published evidence comes mainly from Russian-language journals and from a single research group, with small sample sizes and limited independent replication. A few laboratories outside that group have examined related peptides and reported weaker or absent telomerase effects, so the central claim is best described as contested rather than settled. Rodent studies report modest changes in some ageing markers and in survival, but designs vary and control conditions are often sparse. No large randomised trial in humans has been published, and long-term safety data in healthy populations are correspondingly thin.
Epitalon is a synthetic tetrapeptide with the sequence alanine-glutamate-aspartate-glycine, commonly abbreviated AEDG. It was designed at the St. Petersburg Institute of Bioregulation and Gerontology as a short, chemically defined analogue of epithalamin, a fraction obtained from bovine pineal tissue. Small peptide bioregulators of this type formed a long-running line of work there from the 1980s onward. Because the molecule is produced by solid-phase synthesis rather than extraction, its composition is exact and its purity can be measured directly.
The most frequently cited proposed action is induction of telomerase, the enzyme that adds repeat sequences to chromosome ends. Cell-culture work from the originating group reported higher telomerase reverse transcriptase expression and measurable telomere elongation in human somatic cells after exposure. How a four-residue peptide would reach nuclear gene regulation is not established, and no cell-surface receptor or uptake route has been identified. Additional reports describe changes in melatonin secretion, antioxidant enzyme activity and lipid peroxidation in aged animals, but these findings remain mechanistically unconnected to the telomerase observation.

Identity and purity are usually assessed by reversed-phase high-performance liquid chromatography with ultraviolet detection, frequently near 214 nanometers where the peptide bond absorbs. Mass spectrometry, most often with electrospray ionization, confirms the observed molecular mass and helps separate the intact peptide from truncated or modified forms. Amino acid analysis or tandem mass spectrometry can establish residue order. Purity is commonly quoted as an area percentage, yet such values are method-dependent, and comparisons between laboratories require matching column, gradient and detection wavelength.
Laboratory handling begins with dissolution of the lyophilized powder in water or a suitable aqueous buffer. The dry solid is the more stable form, so stock solutions are generally prepared only when required and kept cold afterwards. Repeated freezing and thawing of a solution is avoided because it encourages aggregation and gradual loss of the intact chain. Diluents and containers are selected to limit adsorption of a short peptide onto plastic surfaces and to reduce microbial growth in aqueous preparations.
Stability depends strongly on pH, temperature and the presence of oxygen and trace metals. Cleavage of the backbone proceeds faster under neutral to alkaline conditions, whereas acidic solutions tend to slow that reaction. The aspartate and glutamate side chains can undergo deamidation or imide formation over time, generating closely related impurities. Published stability data specific to epitalon are sparse, so the usable life of a given solution is best regarded as an open question that depends on buffer composition, concentration and storage temperature.
Residue composition is the property that most cleanly separates verified material from mislabelled samples. Alanine, glutamate, aspartate and glycine appear in that order from the N-terminus, and the two acidic residues sit in the middle of the chain. Because the peptide is short, it can be produced by solid-phase synthesis and characterised by mass spectrometry without ambiguity. Any reported sample whose measured mass departs substantially from 390 daltons is a different compound or a degraded mixture rather than epitalon.
Epitalon is a synthetic tetrapeptide whose four residues are alanine, glutamate, aspartate and glycine, commonly abbreviated AEDG. Its molecular formula is C14H22N4O9 and its monoisotopic mass is near 390.35 daltons. The peptide carries two acidic side chains, so it is neutral to negatively charged in most aqueous buffers. Published reference summaries usually list it under both spellings, epitalon and epithalon, and treat the two names as the same material.
The compound is generally described as a synthetic analogue of a fragment isolated from a pineal gland extract. Researchers associated with the Saint Petersburg Institute of Bioregulation and Gerontology introduced it during the 1980s and 1990s while studying short peptides from animal tissue. The original extract, called epithalamin, is a heterogeneous mixture, whereas epitalon is a single defined sequence. That distinction matters because findings reported for the extract are not automatically findings about the pure tetrapeptide, and claims about broader biological effects remain a separate question from the chemical identity described here.
Human data are limited to small studies, often without the randomization, blinding, or control groups expected in contemporary clinical research. Reported outcomes have included changes in melatonin levels, immune markers, and subjective measures, but sample sizes were generally too small to support firm conclusions. Some reviews treat the peptide as promising while noting methodological weaknesses; others question whether the observed effects are specific. The compound is frequently discussed in longevity-focused communities, where enthusiasm often outpaces the published evidence. Separating established findings from speculation is therefore important when reading summaries of this research.
Epitalon emerged from research conducted in Saint Petersburg by Vladimir Khavinson and colleagues, who studied short peptides as potential regulators of aging. The work built on epithalamin, a pineal gland extract reported to influence neuroendocrine function. Epitalon was designed as a synthetic counterpart with a defined sequence, allowing reproducible experiments that extracts could not support. Early publications described effects on melatonin rhythms and lifespan in animal models. These findings circulated mainly in Russian-language journals during the 1990s, which limited their visibility among English-speaking researchers.
The most widely cited claim is that epitalon activates telomerase and thereby extends telomere length. Supporting evidence comes largely from cultured human cells, where treatment was associated with increased telomerase activity and delayed replicative senescence. Telomerase activation is a biologically consequential effect, since the enzyme is largely silenced in most somatic cells. However, the route by which a short peptide would reach and act on the enzyme's regulatory machinery has not been established. Independent replication in human trials is scarce, so the link between cell-culture observations and whole-body aging remains an open question.
Interest in epitalon is usually discussed within the broader field of short peptide bioregulators, a category that includes other synthetic di-, tri-, and tetrapeptides studied by the same research group. These compounds share a common rationale: that small fragments of tissue-derived proteins can retain biological activity and can be produced reproducibly. The category as a whole remains outside mainstream pharmacological consensus, and epitalon specifically has a limited presence in independent, non-Russian research literature, which shapes how its evidence base is described.
Epitalon is a synthetic tetrapeptide with the amino acid sequence alanine-glutamate-aspartate-glycine, abbreviated Ala-Glu-Asp-Gly or AEDG. It was developed by the Russian researcher Vladimir Khavinson and colleagues during work on peptide bioregulators derived from the pineal gland. The compound is short enough to be produced by standard solid-phase peptide synthesis and is typically handled as a lyophilized white powder. Its small size distinguishes it from larger pineal peptides such as epithalamin, a complex extract from which the tetrapeptide was conceptually derived.
The four residues give epitalon a molecular formula of C14H22N4O9 and a molecular weight near 390.35 daltons. The presence of two acidic residues, glutamate and aspartate, makes the free peptide strongly acidic, while the alanine and glycine ends provide neutral, nonpolar character. This combination produces a molecule with substantial water solubility. Because there are no cysteine, methionine, or tryptophan residues, the peptide lacks the most common oxidation-sensitive side chains, which simplifies handling compared with many longer peptides.
4-HO-DPT acts as a high-efficacy partial agonist to full agonist of the serotonin 5-HT2A, 5-HT2B, and 5-HT2C receptors. It has more than two orders of magnitude greater potency as an agonist of the serotonin 5-HT2A and 5-HT2B receptors than as an agonist of the serotonin 5-HT2C receptor. Hence, it shows considerable selectivity for the serotonin 5-HT2A receptor over the serotonin 5-HT2C receptor. Compared to psilocin (4-HO-DMT), 4-HO-DPT has about the same potency and efficacy as a serotonin 5-HT2A receptor agonist, has about the same potency but is much more efficacious as a serotonin 5-HT2B receptor agonist (EmaxTooltip maximal efficacy = 39% vs. 94%, respectively), and has about the same efficacy but approximately 10-fold lower potency as a serotonin 5-HT2C receptor agonist. 4-HO-DPT produces the head-twitch response, a behavioral proxy of psychedelic effects, in rodents. Its potency for inducing the head-twitch response in mice is about 4- or 5-fold lower than that of psilocin.
MetService issues red-level wind warnings for the Canterbury and Wellington Regions and the Wairarapa south of Carterton. Emergency Management and Recovery Minister Mark Mitchell declares a local state of emergency in Canterbury in response to strong winds and fires. Parliament passes legislation limiting the threshold for Māori foreshore and seabed claims. While government parties supported the legislation, opposition parties vowed to repeal the law change. In response, Te Pāti Māori MPs Debbie Ngarewa-Packer and Tākuta Ferris burnt a copy of the legislation in protest. 23 October – Five unions (the Public Service Association, the New Zealand Educational Institute, the Post Primary Teachers' Association, the New Zealand Nurses Organisation, and the Association of Salaried Medical Specialists) hold a coordinated national strike. Due to severe wind storms nationwide, several protest events in the South Island were cancelled or moved indoors. 24 October: Emergency Management Minister Mark Mitchell declares a state of local emergency for the Southland Region in response to wind and storm damage. The Clutha District Council declares a state of emergency in the Clutha District in response to wind and storm damage. The state of emergency had been lifted for much of Canterbury except the Kaikoura District. Health New Zealand confirms that a recent measles community outbreak was linked to a Northland patient who travelled aboard a Bluebridge ferry across the Cook Strait on 3 October, bringing the total number of measles cases nationwide to eight.
=== Brush type columns (Pirkle Type) === The brush type, or Pirkle type chiral stationary phases are also called π-π Donnor-Acceptor columns. According to some theoretical models separation on these CSPs are based on a three-point attachment between the solute and the bonded chiral ligand on the surface of the stationary phase. These interactions may be attractive or repulsive in nature, depending on the mutual properties. Pirkle columns discriminate enantiomers by binding of one enantiomer with the chiral stationary phase, thereby forming a diastereomeric complex through π-π bonding, hydrogen bonding, steric interactions, and/or dipole stacking. Pirkle CSP can be categorized into three classes: (i) π-electron acceptor (ii) π-electron donor (iii) π-electron donor-π-electron acceptor.
Inhibition of radical SAM enzyme MqnE in menaquinone biosynthesis is reported to be an effective antibacterial strategy against H. pylori. Radical SAM enzyme BlsE has been discovered to be a central enzyme in blasticidin S biosynthetic pathway. Blasticidin S produced by Streptomyces griseochromogenes exhibits strong inhibitory activity against rice blast caused by Pyricularia oryzae Cavara. This compound specifically inhibits protein synthesis in both prokaryotes and eukaryotes through inhibition of peptide bond formation in the ribosome machinery. A new fungal radical SAM enzyme has also been reported to facilitate the biocatalytic routes for synthesis of 3'-deoxy nucleotides/nucleosides. 3'deoxynucleotides are a class of drugs that interfere with the metabolism of nucleotides, and their incorporation into DNA or RNA terminates cell division and replication. This activity explains why this compound is an essential group of antiviral, antibacterial or anticancer drug.
What caused the sizable deficit increases during and shortly after the Great Recession? The CBO reported that the deficit expansion was mainly due to the economic downturn rather than policy choices. Revenue fell while social safety net spending increased for programs such as unemployment compensation and food stamps, as more families qualified for benefits. From 2008 to 2009, the large deficit increase was also driven by spending on stimulus and bailout programs. Should the Bush tax cuts of 2001 and 2003 be allowed to expire in 2010 as scheduled? Ultimately, the Bush tax cuts were allowed to expire for the highest income taxpayers only as part of the American Taxpayer Relief Act of 2012. Should significant deficits be continued or should fiscal austerity be implemented? While the deficit jumped from 2008 to 2009, by 2014 it had fallen to its historical average relative to the size of the economy (GDP). This was due to the recovering economy, which had increased tax revenue. In addition, tax increases were implemented on higher-income taxpayers, while military and non-military discretionary spending were reduced or restrained (sequestered) as part of the Budget Control Act of 2011.

=== Influenza === Flublok Quadrivalent is a licensed recombinant subunit vaccine for active immunisation against influenza. It contains HA proteins of four strains of influenza virus purified and extracted using the Baculovirus-insect expression system. The four viral strains are standardised annually according to United States Public Health Services (USPHS) requirements. Flublok Quadrivalent has a comparable safety profile to traditional trivalent and quadrivalent vaccine equivalents. Flublok is also associated with less local reactions (RR = 0.94, 95% CI 0.90–0.98, three RCTs, FEM, I2 = 0%, low‐ certainty evidence) and higher risk of chills (RR = 1.33, 95% CI 1.03–1.72, three RCTs, FEM, I2 = 14%, low‐certainty evidence).
=== United Cup === Zverev has started his season every year since 2023 by participating in the United Cup. In 2023, Zverev and Team Germany were placed in Group C alongside the Czech Republic and the USA. Against the Czech Republic, he lost to Jiří Lehečka. The Czech Republic won the tie over Germany 3–2. Against the US, he lost to Taylor Fritz. The USA beat Germany 5–0. Germany failed to qualify for the knockout round and ended third in Group C. In 2024, Zverev returned to the United Cup in Sydney, Australia for the second consecutive year, leading Team Germany alongside Angelique Kerber. He won both of his singles matches in the round-robin stage, defeating Lorenzo Sonego of Italy and Adrian Mannarino of France both in three sets. He played both mixed doubles matches in both ties with Angelique Kerber, partnering for the first time since 2019, and won against Team Italy but not Team France. Despite this, Team Germany managed to qualify as the best runner-up team from all countries participating in Sydney, advancing to the quarterfinals against Greece. There, Zverev decisively won his singles match against Stefanos Tsitsipas and his mixed doubles match, partnering with doubles specialist Laura Siegemund, against Maria Sakkari and Petros Tsitsipas. In the semifinal tie against Australia, Zverev was defeated by Alex de Minaur in three sets, allowing the Australian to crack the top 10 after previously making top 10 wins against Taylor Fritz and Novak Djokovic earlier that week.
=== Availability === Gonadorelin is available widely throughout the world for veterinary use, including in the United States, Canada, the United Kingdom, Ireland, elsewhere throughout Europe, South Africa, Australia, New Zealand, Japan, Taiwan. However, gonadorelin is not available for clinical use in humans in the United States.
=== UV-induced DNA damage === UV-irradiation of human skin cells generates damages in DNA through direct photochemical reactions at adjacent thymine or cytosine residues on the same strand of DNA. Cyclobutane pyrimidine dimers formed by two adjacent thymine bases, or by two adjacent cytosine bases, in DNA are the most frequent types of DNA damage induced by UV. Humans, as well as other organisms, are capable of repairing such UV-induced damages by the process of nucleotide excision repair. In humans this repair process protects against skin cancer.
=== Safety concerns and training related to children === Orthopaedic specialists used to recommend that children avoid weight training because the growth plates on their bones might be at risk. The rare reports of growth plate fractures in children who trained with weights occurred under inadequate supervision or improper form or excess weight. There have been no reports of injuries to growth plates in youth training programs that follow established guidelines. The position of the National Strength and Conditioning Association is that strength training is safe for children if properly designed and supervised. The effects of training on youth have been shown to depend on the methods of training being implemented. Studies from the Journal of Strength and Conditioning Research concluded that both Resistance Training and Plyometric training led to significant improvements in peak torque, peak rate of torque development, and jump performance. Plyometric training also showed a greater improvement in jump performance compared to Resistance training. Another study saw results that suggest that both high-load, low-repetition and moderate-load, high-repetition resistance training can be prescribed to improve muscular fitness in untrained adolescents, as well as the jump height had also increased. Both plyometrics exercise and resistance training tend to result in better adaptations in the short and long term. This can be attributed to the effect of neuromuscular development, as well as the principle that it comes faster for adolescents than muscular hypertrophy.

A day after his father's death, the National Assembly received clear instructions to dismiss Natchaba and elect Gnassingbé in his place, which would legalize his succession, which took place on 6 February 2005. Gnassingbé's election was unanimously approved by the deputies (98% of them were members of the ruling party) who were present in the National Assembly at the time; the opposition was not represented in the National Assembly due to its boycott of the 2002 parliamentary election. The members of Gnassingbé's then-party, the Rally of the Togolese People (RPT), did not want to challenge the army's choice. The parliament also eliminated a constitutional requirement that elections be held within 60 days of the president's death, enabling the younger Gnassingbé to rule until the expiration of his father's term in 2008. Under pressure from others in the region, and particularly Nigeria, later that month 2005, Gnassingbé announced that new elections would be held within 60 days, but said that he would remain in office in the meantime. However, on 21 February, the National Assembly reversed some of the constitutional changes that it had made so as to allow Gnassingbé to assume power, although it did not instruct him to resign. This was construed as a way of pressuring him to stand down with dignity. To change the constitution during a period of transition was itself an unconstitutional act, but this did not deter Gnassingbé's allies.
=== Function === Leukotrienes act principally on a subfamily of G protein-coupled receptors. They may also act upon peroxisome proliferator-activated receptors. Leukotrienes are involved in asthmatic and allergic reactions and act to sustain inflammatory reactions. Several leukotriene receptor antagonists such as montelukast and zafirlukast are used to treat asthma. Recent research points to a role of 5-lipoxygenase in cardiovascular and neuropsychiatric illnesses. Leukotrienes are very important agents in the inflammatory response. Some such as LTB4 have a chemotactic effect on migrating neutrophils, and as such help to bring the necessary cells to the tissue. Leukotrienes also have a powerful effect in bronchoconstriction and increase vascular permeability.
The international drug control conventions, also known as the United Nations drug control conventions, are three related, non self-executing treaties that establish an international legal framework for drug control. They serve to maintain a classification system of controlled substances including psychoactive drugs and precursors, to ensure the regulated supply of those substances useful for medical and scientific purposes, and to prevent other uses. Ratification is near universal among UN member countries. The treaties are the Single Convention on Narcotic Drugs (1961; amended in 1972), the Convention on Psychotropic Substances (1971), and the UN Convention Against Illicit Traffic in Narcotic Drugs and Psychotropic Substances (1988). There are also other minor treaties addressing drugs, such as the Convention on the Rights of the Child or the UN Convention on the Law of the Sea.
Depending on the nature of the substance, an elementary entity may be an atom, a molecule, an ion, an ion pair, or a subatomic particle such as a proton. For example, 10 moles of water (a chemical compound) and 10 moles of mercury (a chemical element) contain equal numbers of particles of each substance, with one atom of mercury for each molecule of water, despite the two quantities having different volumes and different masses. The mole is an amount corresponding to a given count (an Avogadro number) of elementary entities. Usually, the entities counted are chemically identical and individually distinct. For example, a solution may contain a certain number of dissolved molecules that are more or less independent of each other. However, the constituent entities in a solid are fixed and bound in a lattice arrangement, yet they may be separable without losing their chemical identity. Thus, the solid is composed of a certain number of moles of such entities. In yet other cases, such as diamond, where the entire crystal is essentially a single molecule, the mole is still used to express the number of atoms bound together, rather than a count of molecules. Thus, common chemical conventions apply to the definition of the constituent entities of a substance, in other cases exact definitions may be specified. The molar mass of a substance is equal to its relative atomic (or molecular) mass multiplied by the molar mass constant, which is almost exactly 1 g/mol.

Reversed-phase high-performance liquid chromatography with ultraviolet detection at 214 nanometres is the usual approach. Peak area percentage provides a purity figure relative to other components in the sample. Mass spectrometry is then applied to confirm identity.
Aspartate can cyclise into a succinimide intermediate that later hydrolyses back as isoaspartate. Both forms share the same molecular mass, so mass spectrometry alone cannot tell them apart. This shapes how purity data should be interpreted.
Freeze-dried material is normally kept desiccated below minus twenty degrees Celsius and shielded from light. Solutions are prepared shortly before use and held cold. Repeated thawing is avoided because it accelerates aggregation and degradation.
No. Epithalamin is a peptide-containing extract of bovine pineal glands, while epitalon is a single synthetic tetrapeptide. The extract contains many peptides and other tissue components, so its composition varies between batches in ways that a synthesised sequence does not.
